V01077必拓生物pTFN载体,pTFN vector,质粒载体图谱
pTFN载体含有分子伴侣FkpA编码序列。大肠杆菌分子伴侣FkpA同时具有肽酰脯氨酰顺反异构酶活性和分子伴侣活性,可以促进外源蛋白的正确折叠,有利于外源蛋白的表达。除此之外,pTFN载体还含有T7启动子,可用于外源蛋白在大肠杆菌体系中的高效表达。载体大小约为6.1kbp,卡钠霉素抗性。

注意事项
1. T7启动子强度较高,为提高可溶表达量,可以适当减少诱导剂的浓度或者降低诱导时的温度;
2.目的蛋白以融合的方式表达在FkpA下游,载体C-末端同时含有His6标签序列,可用于纯化。
