V01043必拓生物pTSN载体,pTSN vector,质粒载体图谱
来自大肠杆菌热稳定的肠毒素II的信号肽(STII)已经被证明是一种高效率的引导外源蛋白分泌的信号肽,依赖于不同的蛋白种类和序列,其分泌产物甚至可以以较高的浓度聚集在培养基中。除此之外,pTSN载体还含有T7启动子,可用于外源蛋白在大肠杆菌体系中的高效分泌表达。载体大小约为5.5kbp,卡钠霉素抗性。

注意事项
1. T7启动子强度较高,为提高分泌量,可以减少诱导剂的浓度至0.05 mM IPTG;
2.可以采用自诱导的方式分泌表达目的蛋白;
3.不同种类或不同序列的蛋白,其分泌效率差异很大,需要进行预实验确定较好的条件
