Background:

Suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. Regulates cell death by controlling the mitochondrial membrane permeability. Appears to function in a feedback loop system with caspases. Inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1).

Assay Conditions:

Following the protocol for BPS Cat. #50222. Briefly, BCL-2 was combined with BCL-2 peptide ligand in TR-FRET assay buffer along with anti-His Tb donor and dyelabeled acceptor. Varying concentrations of ABT 199 were added and the reaction was incubated at room temperature for 2 hours followed by fluorescent intensity measurements.

40 mM Tris-HCl, pH 8.0, 110 mM NaCl, 2.2 mM KCl, 3 mM DTT, 20% glycerol, and 200 mM imidazole
